PT-141

PT-141 (bremelanotide) is a synthetic peptide derived from alpha-melanocyte-stimulating hormone. It acts on melanocortin receptors, particularly MC4R, and has been studied for hypoactive sexual desire disorder in both men and women.


It is primarily researched for its effects on sexual arousal pathways and has also been investigated for other physiological roles related to melanocortin signaling, though its main clinical focus remains sexual function.


Sexual Arousal

PT-141 works by activating melanocortin receptors, especially MC-4R, which are involved in sexual arousal and behavior. Unlike drugs such as Viagra, it acts mainly through the central nervous system rather than directly improving blood flow to the genitals.


Studies have shown that PT-141 may help some men with erectile dysfunction who do not respond to sildenafil. Research has also explored its potential use for female sexual dysfunction and hypoactive sexual desire disorder (HSDD). The newer formulation, Vyleesi, became available in the United States in 2019 for certain cases of HSDD in premenopausal women.


PT-141 has also been studied as a possible treatment for sexual dysfunction in both men and women. However, its effectiveness, safety, and appropriate medical use depend on the condition being treated and require professional guidance.

Hemorrhage

A modified form of PT-141, called PL-6983, was investigated as a possible treatment for hemorrhagic shock. By acting on MC-1R and MC-4R, it may help reduce tissue damage caused by low blood flow during severe blood loss. The compound reached Phase IIb clinical testing.


Infection

Research in rats suggests that MC-1R may have anti-fungal and anti-inflammatory effects. These findings have encouraged interest in melanocortin-based treatments as possible alternatives for certain fungal infections, especially when current treatments have limitations or cause significant side effects.


Cancer

MC-1R is involved in inflammation and DNA repair, making it a subject of cancer research. Certain MC-1R variations have been linked to an increased risk of basal cell and squamous cell skin cancers. Researchers are investigating whether modified peptides related to PT-141 could influence these pathways and potentially help prevent or treat certain cancers.
